General and Technical Questions
Q: Can I order free saccharides?
A: Usually free saccharides and aminoalkyl glycosides are not available for sale. Nevertheless, some of them could be available on custom synthesis principle. Please contact us directly by tel/fax: 7 (495) 330 55 92 or e-mail: atanov@carb.ibch.ru
Q: What is composition of conjugates?
A: We offer to our customers the conjugates based on polyacrylamide loaded with 20% mol. of oligosaccharide, in accordance with our experience this value is optimal. Carbohydrate probes additionally contain the biotin (5 %mol.), fluorescein (1 %mol.), or other label.
The knowledge of the optimal conditions for the condensation reaction gives us the possibility to synthesize conjugates with the exactly defined molar content of ligands. The real size of the conjugate was evaluated by electron microscopy: the molecule had an almost perfectly spherical form with the diameter about 150Å, which agrees well with its hydrodynamic size by indirect methods. For the majority of bioanalytical problems, the molecular mass of conjugates of about 30-50 kDa is sufficient and probably optimal.
More information about composition, properties, labels see in Review "Polyacrylamide-Based Glycoconjugates as Tools in Glycobiology" [Glycoconjugate Journal, 15, 431-446, 1998].
Q: What is OPTI-SET ?
A: We produce series of unlabelled PAA-based neoglycoconjugates with different content of carbohydrates (for example: 2, 5, 10, 15, 20 and 30% mol.) to optimize studying of their interaction with carbohydrate-binding materials. In case you choose the optimal conjugate for your assay from SET you could order it separately, price of it shall correspond to regular 20% conjugate of the same ligand. Labelled SETs may be enquired too.
Q: What is the purity of ordered product?
A: Synthetic w-aminoalkyl glycosides used for coupling with the polymer have a purity of >95% as controlled by HPLC and NMR. Composition of the conjugates is confirmed by acid hydrolysis followed by HPLC of AMC-monosaccharides (also see chart "Quality Control"). We have specifications which describes data analysis, storage and other information. Please note in the order if you wish to receive enclosed certificate of analysis.
Q: Can I order a specific lot?
A: You can request a definite lot you have used before, but we cannot guarantee it will still be in stock. When you place the order, please note if you would like to reserve more quantity of certain reagents.
Q: How to keep reagents?
A: Storage: long-term storage for sialylated, phosphated and sulfated compounds we recommend at -20°C, for other probes at 4°C. Concentrated solutions of the conjugates can be stored for several days in glassware at 4°C, for long-term storage we recommend to lyophilize the solutions again. Long-term storage of diluted solutions of glycoconjugates in plastic ware causes the decrease of it's concentration due to adsorption on vessel surface.
Q: Do you have a protocol for the use of this product?
A: Many of our products come with use instructions, and almost all of our products come with a list of related articles. Even when we have no specific information, however, we may be able to direct you to references or protocols for related products. Please send your questions by fax: 7 (495) 330 55 92 or e-mail: atanov@carb.ibch.ru
Q: What do you recommend as negative control?
A: We offer to our customers to use corresponding derivatives of aminoglucitol HOCH2(HOCH)4CH2NH-(ligand # 00) as negative control. However in each case you can choose additionally more suitable negative controls, for example during analyses of anti-Ley antibodies specificity you can use Lex-PAA, H(type 2)-PAA and Leb-PAA.
Q: What glycoadsorbents do you have?
A:
We have adsorbents based on rigid macro porous glass particles (pore diameter 2000Å) or on Sepharose FF. Both of them have macro porous structure of the carrier enables its high capacity.
We have listed here adsorbents in our stock. Adsorbents loaded with other carbohydrate ligands manufactured by We are available on custom synthesis principle.
We also is ready to prepare adsorbents from aminoalkyl glycosides, glycopeptides or reducing saccharides provided by the customer.
Q: ELISA procedure.
A: The typical description of ELISA with the use of We's neoglycoconjugates see in the section "ELISA and ELLA".
